Yes, the USB port can always be used for packet serial commands, whatever the mode of the board is set to.
Two limitations. If the mode of the board is set to something other than packet serial mode then the other mode has priority for driving the motor(eg RC mode will prevent motor movement commands from doing anything when a packet serial motor movement command is set through USB). Second, you will most likely need to use a USB cable with a RF choke near the Roboclaws USB connection. Noise from the motors can get back to the computer if you dont which can cause the USB to think an error has accured and close the port.
